EM Sample Preparation Workshop

The key apparatuses used to prepare sample with cryo-TEM technology include high pressure freezing apparatus (Leica EM HPM100, Leica EM PACT2), cryosectioning apparatus (Leica EM UC6, Leica EM FC6, Leica EM FC7), and freeze substitution apparatus (Leica AFS). With these apparatuses, we can prepare biology samples with high pressure rapid freezing fixation, cryosectioning and cryo-dehydrating-embedding-polymerizing methods. Combining with cryo-TEM technology, the natural order structure of biological sample can be observed, and experiments of immuno-TEM and 3D-structure reconstruction can be done.

 

Leica EM HPM100

The Leica EM HPM100 is a high pressure freezing system for freezing aqueous specimens order to be examined with TEM,SEM, AFM and LM. It makes it possible to observe aqueous biological and industrial samples near to native state. Specimens are simultaneously pressurized (210kPa) and frozen with liquid nitrogen at a temperature of -196°C.

The Leica EM HPM100 can vitrifies samples up to 200µm in thickness. The 3mm and 6 mm diameter carrier system of the Leica EM HPM100 allows more sample area to be frozen. The Fast sample loading design of the Leica EM HPM100 provides fast sample loading, automatic freezing of samples with one button operation and automatic sample transfer to liquid nitrogen. It enables express sample handling and easy use with good freezing results.  

  

 

Leica EM PACT2

The Leica EM PACT2 is a high pressure freezing system for vitrifying samples up to 200µm in thickness without the artefacts of chemical fixation. The Leica EM PACT2 makes it possible to observe aqueous biological and industrial samples near to native state. Liquid nitrogen be used as the cryogen. The Leica EM RTS rapid transfer system allows correlative LM/EM experiments, taking a live specimen from light microscopes (e.g. confocal) to freezing in less than 5 seconds.

The Leica EM PACT2 has an Automatic specimen unloading device into the LN2 bath ensures that the sample does not thaw after freezing; no crystallization contamination occurs and the specimen is protected. In the freezing, a specimen holder containing sample is placed in the chamber. Approximately 20ms after reaching 2000 bar a jet of LN2 is applied onto the outer surface of the sample holder. After freezing, the specimen holder is automatically ejected into an LN2 bath.  

  

 

Leica EM UC6

The Ultramicrotome Leica EM UC6 provides preparation of semi- and ultrathin sections of biological and industrial samples for TEM, SEM, AFM and LM examination.

The  Leica EM UC6 has three independent brightness-controlled LED light sources for toplight, backlight and specimen transillumination. The cutting window is from 0.2 to 15mm.The cutting speed can be set in a wide range from 0.05 to 100 mm/sec. Section thickness setting from 1nm to 2.5um and semithin from 2.5um to 15um.  

 

 

Leica EM FC6

The Cryo-ultramicrotome Leica EM FC6 is the low temperature sectioning system based on the Ultramicrotome Leica EM UC7 by mounting a Cryochamber. It provides preparation of semi- and ultrathin cryo-sections (-15° to -185°C) of biological and industrial samples for TEM, SEM, AFM and LM examination.

For biological samples, The Leica EM FC6 is used for cryoprotected sectioning (Tokuyasutechniques) and Frozen Hydrated Sectioning (FHS). With the integrated antistatic device it provides good cryo-sectioning performance.  

 

 

Leica EM FC 7

The Cryo-ultramicrotome Leica EM FC7 is the low temperature sectioning system based on the Ultramicrotome Leica EM UC7 by mounting a Cryochamber. It is used for semi- and ultrathin cryosectioning of biological and industrial samples for TEM, SEM, AFM and LM examination. This includes Frozen Hydrated Sectioning (FHS) of vitrified specimens, cryoprotected biological samples (Tokuyasu techniques) and soft industrial specimens.

The Leica EM FC7 has an integrated chamber illumination, allows to use an integrated antistatic device and provides a wide temperature range from -15° to -185°C for cryosectioning. The heated chamber walls prevent icing during extended working time. Especially for frozen hydrated sectioning or the Tokuyasu method, the Leica EM CRION with electrostatic discharge and charge mode used with the micromanipulator provides good cryo-sectioning performance.  

 

 

Leica AFS

The Leica EM AFS2 performs freeze substitution and progressive lowering of temperature (PLT) techniques and allows low temperature embedding and polymerization of resins.

The Leica EM FSP (freeze substitution processor), an automatic reagent handling system combined with the Leica EM AFS2, dispenses reagents for both freeze substitution and PLT applications. The LED illumination from within the chamber and the attached stereomicroscope for viewing and positioning of samples ensures ease of use.
